Automated
The
Parallab® 350 is designed for true unattended
automation and walk away processing. The system stores
all of the components for the reactions and cleanup (e.g. template,
Big Dye® cocktail, magnetic beads for purification, alcohol
and water) for a full day's run on a chilled work deck.
During
the reaction assembly, the robot moves to the required plate location
and removes the insulated lid to provide access for the Nano-Pipetter.
The Nano-Pipetter aspirates a small volume of reagent
into each of its 96 thin-walled glass capillaries, each serving
as a reaction vessel.
After all of the components are automatically drawn into the Nano-Pipetter,
they are effectively mixed using a high speed shuttling of the fluids
inside the capillaries. The Nano-Pipetter is then
seated in the onboard high speed thermal cycler, where the 96 capillaries
are reversibly sealed and thermal cycled. Following thermal
cycling, magnetic bead solution is aspirated into each reaction
tube where the purification takes place. The 96 cleaned up, final
products are then dispensed into an output plate for off-line
analysis. The Nano-Pipetter is then automatically
cleaned with the onboard wash station in preparation for the next
run.
